Human lymphocytes can be categorized into three primary subset populations based on their biological functions and cell-surface antigen expression: T lymphocytes (CD3⁺), B lymphocytes (CD19⁺), and NK lymphocytes
(CD16⁺CD56⁺). The Cenglang™ 6-Color TBNK reagent is designed to characterize these lymphocyte subset populations with or without the use of counting beads.
When used in conjunction with counting beads, compatible cytometry software calculates lymphocyte subset percentages and absolute counts directly from flow data obtained from both the beads and the sample. In the absence of counting beads, the software determines lymphocyte subset percentages directly from sample flow data. If absolute count data from an alternative instrument or method are provided, the software can also derive absolute counts for each subset. Furthermore, Challenbio flow cytometers enable direct absolute counting of samples via the volumetric method, eliminating the need for counting beads.
Quantifying CD3⁺CD4⁺ lymphocyte percentages or counts is particularly valuable for monitoring human immunodeficiency virus (HIV) infection. Individuals with HIV typically exhibit a progressive decline in CD3⁺CD4⁺ lymphocyte counts as the disease advances. Additionally, CD3⁺CD4⁺ percentages or counts, along with total T and B lymphocyte measurements, are instrumental in characterizing and monitoring various forms of immunodeficiency and autoimmune diseases.
NK lymphocytes, identified as CD3⁻CD16⁺ and/or CD56⁺, have been demonstrated to mediate cytotoxicity against specific tumors and virus-infected cells. Notably, NK-mediated cytotoxicity does not require the presence of class I or class II major histocompatibility complex (MHC) molecules on the target cell.
